Abstract

Parijoto fruit (Medinilla speciosa Blume) is traditionally used to support reproductive health, but its safety during pregnancy requires scientific evaluation. This study evaluated the teratogenic potential of parijoto fruit methanol extract in pregnant ddY mice. Forty-four pregnant mice were allocated to vehicle control and treatment groups, with 22 dams per group. The control group received 1% sodium carboxymethyl cellulose containing Tween 60, whereas the treatment group received the extract orally at 1000 mg/kg body weight/day on gestation days 5-15. On gestation day 18, laparotomy was performed, and maternal clinical condition, body-weight change, fetal viability, sex, body weight, body length, tail length, and external morphology were assessed using the litter as the analytical unit. No treatment-related behavioral changes, maternal clinical toxicity, fetal deaths, or external morphological malformations were observed. Maternal body-weight gain was significantly greater in the treatment group (p = 0.007). All 375 fetuses were alive, comprising 179 fetuses in the control group and 196 in the treatment group. The number of live fetuses per dam and mean fetal body weight per litter were significantly higher in the treatment group (p = 0.049 and p = 0.001, respectively). Fetal body length, tail length, and sex distribution did not differ significantly between groups (p = 0.769, p = 0.985, and p = 0.922, respectively). Under the study conditions, the extract did not produce adverse macroscopic teratogenic effects. However, comprehensive prenatal safety could not be established because implantation, resorption, visceral, and skeletal endpoints were not evaluated.
