Abstract

Moringa oleifera leaves has bioactive flavonoid compounds that possessed pharmacological activity. However, they face limitations such as low solubility, and poor bioavailability. The development of a nanoparticle-based (AgNP) drug delivery system is expected to address these issues. Silver nanoparticles tend to aggregate and lack stability. Incorporating a stabilizing agent, such as chitosan, into the nanoparticles offers a potential solution to enhance the stability and applicability of AgNPs. This study aims to characterize the physicochemical properties of silver nanoparticles synthesized using Moringa oleifera leaf extract, both without and with chitosan encapsulation (AgNP-Chitosan). The research involved extracting Moringa leaves via maceration, determining total flavonoid content, biosynthesizing AgNPs through a green synthesis method (reducing 1 mM AgNO₃), and encapsulating them with 0.2% chitosan. Nanoparticle characterization parameters included particle size, polydispersity index, zeta potential, and entrapment efficiency. Characterization results showed particle sizes of 20.8 ± 4.7 nm (AgNP) and 12.6 ± 2.8 nm (AgNP-Chitosan); polydispersity index values of 0.787 (AgNP) and 0.732 (AgNP-Chitosan); zeta potential values of -12.6166 ± 3.5701 mV (AgNP) and -16.8333 ± 4.2697 mV (AgNP-Chitosan); and entrapment efficiencies of 16.80 ± 0.23% (AgNP) and 15.23 ± 1.75% (AgNP-Chitosan). Statistical analysis revealed no significant differences (p > 0.05) in the physical characteristics of AgNPs and AgNP-Chitosan, leading to the conclusion that the addition of chitosan did not significantly affect the silver nanoparticles derived from Moringa leaf extract.
