Abstract
Backgrounds Antioxidants are originate from endogenous sources or exogenous sources like animals, plants, and endophytic bacteria. Moringa oleifera L. possesses high antioxidant activity, yet direct utilization requires large quantities and time. Symbiotic endophytic bacteria offer an alternative to produce similar bioactive compounds rapidly without damaging the host. This study aims to determine the antioxidant activity of metabolites from Moringa leaf endophytic bacteria extracted using Ultrasonic Assisted Extraction (UAE). Methods This in vitro experimental study involved leaf sterilization, isolation on Nutrient Agar, colony screening via catalase tests, fermentation in Nutrient Broth, and extraction using UAE at 40 kHz and 45°C for 20 minutes. Antioxidant activity was tested using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method to determine the Inhibitory Concentration (IC50) value. Results Five bacterial colonies were obtained, with colony number 5 showing the highest catalase activity. Macroscopic identification revealed white colonies with smooth margins and a flat elevation. Microscopic identification revealed bacillus-shaped bacteria, arranged in chains, which were Gram-positive. The antioxidant activity test of the metabolite extract yielded an average IC50 value of 111.436 ppm. Based on these results, the antioxidant activity of Moringa leaf endophytic bacterial metabolites is categorized as moderate. Conclusion The study demonstrated an increase in antioxidant activity to a moderate level in the extract of Moringa leaf endophytic bacterial compounds obtained via the UAE method.
