Abstract

Irritant contact dermatitis (ICD) is a inflammatory skin disease, happened about 20% population worldwide and mostly in occupational settings, that may lead to lower quality of life and job loss. Yet, gel corticosteroid as ICD treatment has concerning adverse effects. Graptophyllum pictum (L.) Griff or daun ungu is a common medical plant in Indonesia that has been studied to possess many promising medical properties, including anti-inflammation, and thus serves as a promising novel topical anti-inflammatory drug to treat ICD. This in vivo study aims to evaluate the anti-inflammatory activity of the gel Graptophyllum pictum (L.) Griff (GPG) extract in a xylene-induced rat-ear edema model. Six male Sprague-Dawley Rats randomly divided into three groups of GPG dosage (150, 300, and 450 mg/kgBW) and compared to control group of Diclofenac gel (DG). The degree of ear thickness and ear weight were evaluated and analyzed statistically. This study demonstrates a different trend in ear thickness change with dose-dependent manner. GPG 450 mg/kgBW group showed a similar change with DG group and displayed the most significant anti-inflammatory activity compared to control group. Therefore, GPG, especially in 450 mg/kgBW dose, exhibited an effective ear edema attenuation in xylene-induce ear edema model and serves a potential novel anti-inflammatory drug. Further molecular mechanism and clinical studies need to investigated.
