Abstract
Antibiotic resistance against Escherichia coli and Staphylococcus aureus has become a major obstacle in the effectiveness of treating infections, especially as amoxicillin, which is a first-line therapy, is now showing high levels of resistance. One strategy to overcome this resistance issue is to combine antibiotics with natural substances that have potential as antibacterial agents, such as sappan wood (Caesalpinia sappan L.). This study aims to evaluate the inhibitory effect of the combination of ethanol extract of sappan wood (Caesalpinia sappan L.) with amoxicillin against Escherichia coli and Staphylococcus aureus. The extraction of sappan wood was carried out using the maceration method with 96% ethanol as the solvent. Phytochemical screening results show the presence of flavonoid, alkaloid, tannin, saponin, phenol, and triterpenoid compounds in the extract. This study evaluates the inhibitory effect using the disc diffusion method with data interpretation using AZDAST. The interaction of the combinations showed a synergistic effect against Staphylococcus aureus at concentrations of 2,000-5,000 ppm, whereas it showed an antagonistic effect on Escherichia coli at all tested concentrations. Data analysis using Kruskal Wallis and Mann Whitney indicated tests indicated statistically significant differences in the diameter of the inhibition zones between treatment groups (p < 0.05).
