Abstract
Hyperglycaemia with increased free radicals affects organ quality down to the cellular level. Oxidative stress triggers apoptosis, involving anti-apoptotic (Bcl-2) and pro- apoptotic (BAX) genes. Spermatogenesis disruption, manifesting as decreased concentration and morphological abnormalities, may result from excessive apoptosis during mitosis and meiosis. The impact of flavonoids in Moringa oleifera on spermatogenesis has proven to enhance sperm count and male reproductive system function. The research adopts true experimental design, analysing BAX gene expression, concentration, and sperm morphology in hyperglycaemic white rats treated with Moringa oleifera leaf extract, comparing treatment and control groups. Gene expression is quantified relative to the comparative threshold cycle (CT) using RT-PCR. Comparing fold change with the baseline of 1 in the negative control group, treatment groups exhibit values approaching this figure. The highest mean sperm concentration is 66 x 106/ml in the treatment 3 group (400 mg/KgBW), with 94.5% normal sperm morphology. The study concludes that higher extract doses correlate with decreased BAX gene expression and improved white rat sperm quality.
