Abstract
The Moringa plant (Moringa oleifera L.) has many nutritional benefits and pharmacological activities, especially due to the quercetin content found in its leaves. This research aimed to determine the optimal eluent ratio from a combination of methanol and aquabides for testing quercetin levels in dry Moringa leaf extract samples. Additionally, the analytical method was validated to ensure it met the requirements for linearity, precision, accuracy, specificity, and range. The research method involved varying the eluent composition of methanol at ratios of 60:40, 65:35, and 70:30 using a High Performances Liquid Chromatography (HPLC) instrument. The results showed that the optimal eluent ratio was methanol:aquabides (60:40), based on parameters such as retention time (tR), tailing factor (As), area (uAu), and capacity factor (k). The analytical method validation results met the requirements set out in ICH Q2 (R1), with a retention time (tR) value of 2.25 minutes, follow-up factor (As) of 0.83, peak area of 28,764 ± 159.7, and capacity factor (k) of 7.81 ± 0.11. Based on this research, it can be concluded that the optimal eluent ratio is 60:40, the method used has met the validation requirements, and the dry extract of Moringa leaves obtained through the Heat-assisted Extraction process at a temperature of 60°C with 96% ethanol solvent contains quercetin at (22.68 ± 0.30) %.
