Abstract

Polyethylene is the most widely produced type of plastic and is a major contributor to microplastic pollution. Microplastics can enter the body through oral, inhalation, and dermal routes and may adversely affect the reproductive system, particularly sperm viability, which is highly susceptible to oxidative stress. This study aimed to analyze the effect of short-term polyethylene microplastic exposure on sperm viability in male Wistar rats (Rattus norvegicus). This study was a true experimental study with a post-test only control group design using 28 male Wistar rats that were randomly divided into four groups (n=7), namely a negative control group and three treatment groups administered Low-Density Polyethylene (LDPE) microplastics at doses of 0.0375, 0.3, and 0.6 mg/day orally for 14 days. Sperm viability was assessed using eosin staining. The data were analyzed using the Kruskal–Wallis test followed by the Mann–Whitney post hoc test. Mean sperm viability decreased with increasing doses of polyethylene microplastics, measuring 75.3%, 57.9%, 44.2%, and 36.4% in the control, low-, medium-, and high-dose groups, respectively. The Kruskal–Wallis test showed a significant difference among the groups (p=0.001), while the Mann–Whitney post hoc test demonstrated that all treatment groups differed significantly from the control group (p<0.05).Short-term exposure to polyethylene microplastics significantly reduced sperm viability in male Wistar rats (Rattus norvegicus) in a dose-dependent manner.
