Abstract

Red spinach (Amaranthus tricolor L.) contains betalain compounds that have the potential to promote hair growth and can be formulated into a hair tonic as a form of scalp and hair care. This study aimed to determine the betalain content and to analyze the effect of hair tonic formulations containing the betalain fraction of red spinach leaves on hair growth in New Zealand White rabbits. Extraction was performed using ultrasonic-assisted extraction (UAE), followed by liquid-liquid fractionation using n-hexane, ethyl acetate, and ethanol. Total betalain content was determined using UV-Vis spectrophotometry, and the fraction with the highest betalain content was formulated into hair tonic at varying concentrations of 1% (F1), 2% (F2), and 3% (F3). Hair growth efficacy was assessed over 21 days by measuring hair length and weight, and the data were analyzed statistically using comparative tests (ANOVA/Kruskal-Wallis) and correlation tests (Spearman/Pearson). The results showed that the highest betalain content was found in the ethanol fraction (2,327.635 mg/L). The F3 formulation, containing 1,566.33 mg/L of betalain, produced the highest hair length (16.28 mm) and weight (258.06 mg) among the three formulas on day 21. However, statistical analysis revealed no significant differences in hair length (p>0.05), whereas hair weight showed a significant difference (p=0.044) with a weak correlation (r=0.352; p=0.128). Overall, variations in betalain fraction concentration (1–3%) demonstrated a weak and non-significant correlation with hair growth, although the 3% concentration provided the most optimal response, indicating the need for further studies to obtain a more effective formulation
