Abstract
Biofilm produced by cariogenic microorganism, including Streptococcus mutans and Candida albicans, play a crucial role in the development of dental caries and contribute to greater resistance againts antimicrobial treatments. This condition emphasizes of effective natural antibiofilm agents as well as the application of eco-friendly solvents. Butterfly pea flower (Clitoria ternatea L.) has anthocyanins and other secondary metabolites with potential as natural antibiofilm. This study aimed to compare the antibiofilm activity of ethanol extract and NADES extract of butterfly pea flower against S. mutans and C. albicans. Extraction was performed using the Ultrasound-Assisted Extraction (UAE) method with 70% ethanol and NADES choline chloride–glycerol as solvents. Total anthocyanin content was quantified using the pH differential method, while MIC and antibiofilm activity were evaluated using the microdilution method. The results showed that both ethanol and NADES extracts significantly reduced biofilm formation when compared with the negatiive control, with IC50 values under 50%. Both extracts had MIC values at a concentration of 50% against both microorganisms. The NADES extract showed a higher total anthocyanin content than the ethanol extract. Although the IC50 value of the NADES extract tended to be lower, statistical analysis indicated no significant difference between the antibiofilm activities of the ethanol and NADES extracts. This study concludes that NADES has potential as an environmentally friendly alternative solvent for butterfly pea flower extraction that effectively supports the inhibition of biofilm formation by S. mutans and C. albicans.
