Abstract

ABSTRACT Candidiasis caused by Candida albicans is a global health concern worsened by increasing resistance to conventional antifungals such as nystatin. Lemongrass (Cymbopogon citratus) contains active compounds citral, flavonoids, saponins, and tannins that show potential as alternative antifungal agents. Ultrasonic-Assisted Extraction (UAE) was employed to optimize bioactive compound extraction while preserving the quality and stability of thermolabile components such as citral. This study aimed to determine the inhibitory activity, compare antifungal effectiveness across concentrations, and identify the most effective concentration of lemongrass extract obtained by UAE against C. albicans in vitro. A true experimental study with a post-test only control group design was conducted. Lemongrass extract was obtained using UAE with 96% ethanol. Antifungal effectiveness was assessed using the well diffusion method on Sabouraud Dextrose Agar (SDA) incubated for 24 and 48 hours at 37°C. Four treatment groups (10%, 15%, 20%, and 30% concentrations), a positive control (Nystatin 1,000 IU/mL), and a negative control (sterile distilled water) were each replicated four times. Data were analyzed using Kruskal-Wallis with Mann-Whitney on 24-hour observation, and One-Way ANOVA with Tukey HSD on 48-hour observation with the results that all extract concentrations produced inhibition zones against C. albicans. Lemongrass extract (Cymbopogon citratus) obtained by UAE demonstrated antifungal activity against C. albicans. The 30% concentration was the most effective, achieving strong inhibition. A consistent dose-response relationship was observed between increasing extract concentration and increasing inhibition zone diameter.
