Abstract
Objective: Obesity is recognized as a chronic low-grade inflammatory condition characterized by increased levels of Tumor Necrosis Factor-α (TNF-α). This proinflammatory cytokine plays a role in insulin resistance and metabolic diseases. Moringa oleifera fruit extract is rich in bioactive anti-inflammatory compounds that are presumed to suppress TNF-α levels. This study aimed to evaluate the effect of Moringa oleifera fruit extract on TNF-α levels in Sprague–Dawley rats with experimentally induced chronic obesity. Methods: This study employed a true experimental design with a post-test only control group approach. Obesity was induced in 30 male rats through a high-fat diet for 49 days. The treatment groups consisted of a normal control, negative control, positive control, and groups receiving Moringa oleifera fruit extract at a dose of 500 mg/kg body weight with varying administration frequencies (once and twice daily). After a two-week intervention period, TNF-α levels were measured using the ELISA method. Results: ANOVA analysis showed a significant difference among groups (p<0.001). Post hoc Tukey analysis revealed a significant difference between treatment group 1 and treatment group 2 (p<0.001), with a significant reduction in TNF-α levels in the treatment groups compared with the negative and positive control groups. No significant difference was observed between once-daily and twice-daily administration (p>0.05). However, TNF-α levels in the treatment groups were lower than those in the vitamin C group. Conclusion: It can be concluded that Moringa oleifera fruit extract is effective in reducing TNF-α levels. Since no significant difference was found between once-daily and twice-daily administration (p>0.05), once-daily administration is considered an effective dose for reducing TNF-α levels.
