Abstract
The moringa leaf has been found to possess antibacterial activity attributed to active compounds such as flavonoids. Flavonoids offer numerous benefits to the human body, one of which is antibacterial activity. Escherichia coli is among the bacteria commonly found in humans. Antibacterial testing can be performed using diffusion methods, namely disc diffusion and well diffusion. Both methods have their respective advantages, prompting researchers to explore further into comparing disc diffusion and agar diffusion methods for antibacterial testing to clearly observe differences in inhibition zones. Moringa extract was obtained using ultrasonic extraction, and it contains flavonoid levels of 24.67 mg QE/g ± 6.2668. Concentrations of moringa leaf extract used for the antibacterial comparison were 5%, 10%, 20%, 40%, and 80%. The results of this study indicate that the agar diffusion method yields larger inhibition zones compared to the disc diffusion method. This is evidenced by the largest inhibition zone of 23.05 mm observed with an 80% extract concentration using the agar diffusion method. In contrast, the largest inhibition zone observed with the disc diffusion method was only 15.31 mm, also with an 80% extract concentration. Based on Kruskal-Wallis statistical tests, significant differences in inhibition zones were found across several concentrations of moringa leaf extract in both methods (p<0.05).
