Abstract

Acne is a disease that has dominated many populations globally, caused by the bacterium Propionibacterium acnes which has shown resistance to several antibiotics. Zanthoxylum acanthopodium DC. fruit, traditionally used as a spice in Indonesia, has potential antibacterial properties. This study evaluates the antibacterial activity of ethanolic extract of Andaliman fruit against Propionibacterium acnes using the microdilution method. Extraction was performed using Ultrasonic Assisted Extraction at frequencies (30 kHz, 40 kHz, and 50 kHz) and durations (10, 15, and 20 minutes). Phytochemical screening indicated the presence of flavonoids, alkaloids, tannins, steroids, and saponins in the extract. The results showed antibacterial activity against Propionibacterium acnes, with the Minimum Inhibitory Concentration (MIC) observed at 5000 ppm for extracts at 40 kHz frequency and extraction durations of 10 and 15 minutes. However, the Minimum Bactericidal Concentration (MBC) test yielded negative results, indicating no bactericidal effect. Kruskal-Wallis and Dunn tests in SPSS showed significant differences between extraction frequency and duration compared to the positive control, and significant differences among groups in MIC and MBC values (p < 0.05).
