Abstract

Cigarettes can reduce spermatozoa motility by increasing levels of Reactive Oxygen Species (ROS). ROS can be reduced with antioxidants. Flavonoid compounds, especially n-butanol, is one of the antioxidants found in the roots of qustul hindi which is currently widely consumed by people in Indonesia. The aim of this research was to determine the effect of Qustul Hindi root extract on the motility of rat spermatozoa that had been exposed to cigarette smoke. This experimental research was carried out at iRATco Veterinary Laboratory Services Bogor in May-July 2023. The subjects used were male rats (Rattus norvegicus) of the Wistar strain. The inclusion criteria for research samples were healthy and active white Wistar rats, aged 8-10 weeks with a body weight of 150-200 grams. Mice with anatomical abnormalities and those that had been used for previous experiments were excluded. Rats that met the criteria were then divided into five groups, K1, K2 (exposed to cigarette smoke), K3 (exposed to cigarette smoke and given qustul Hindi at a dose of 50 mg/kgBW/day), K4 (exposed to cigarette smoke and given qustul hindi dose 100 mg/kgBW/day) and K5 (exposed to cigarette smoke given qustul hindi dose 200 mg/kgBW/day). Bivariate testing using One Way Anova found p 0.002 which shows that there is an effect of giving Qustul Hindi root extract on spermatozoa motility. In post hoc testing LSD shows significant differences between K1 and K2 & K4, K2 and K3 & K5, K3 and K4, K4 and K5. Administration of Qustul Hindi root extract at doses of 50 and 100 mg/kgBW/day can increase spermatozoa motility, while a dose of 200 mg/kgBW/day can reduce spermatozoa motility.
