Abstract
Introduction Shigella dysenteriae is a Gram-negative bacterium that causes dysentery. Dysentery is generally treated with antibiotics; however, there is an increasing incidence of antibiotic resistance in S. dysenteriae. Actinomycetes are currently recognized as potential microorganisms because they can produce antibacterial agents. Ultrasound-Assisted Extraction (UAE) is modern extraction methods that is widely used today. UAE can significantly shorten the extraction time and improve both the yield and quality of the extract. Several factors affect extraction results, including frequency. This study aims to determine the effect of different UAE frequencies on the effectiveness of Actinomycetes isolates as antibacterials in inhibiting the growth of S. dysenteriae. Method The method applied in this study was a pure experimental design. The antibacterial activity test used the well-diffusion method, and the inhibition potential of the Actinomycetes extract was measured based on the diameter of the inhibition zone formed after ultrasonic extraction at frequencies 30 kHz, 40 kHz, and 50 kHz. Result The results of this study showed the average diameters of the inhibition zones at extraction frequencies of 30 kHz, 40 kHz, and 50 kHz were 3,02 mm; 3,60 mm; and 1,62 mm. Data analysis using the Kruskal-Wallis and Mann-Whitney tests revealed significant differences in antibacterial inhibition among the treatment groups. Discussion The results, the different frequencies of UAE affect the effectiveness of actinomycete isolates in inhibiting the growth of S. dysenteriae. Actinomycetes effectively inhibit the growth of bacteria by targeting the peptidoglycan in the bacterial cell wall, which causes cell death.
