Abstract

Adipose mesenchymal stem cells (AMSCs) are multipotent stem cells with potency for tissue therapy. Isolated primary mesenchymal stem cells are low in numbers, so in vitro expansion in culture medium is needed. DMEM and fetal bovine serum (FBS) are required for the medium. When used for AMSCs culture supplement, FBS has its disadvantages, one of them is the expression of Neu5Gc xenoantigen, which is harmful for human. Honey and royal jelly are known to have the potential composition needed as an alternative of FBS. This study aimed to determine the effect of serum-free DMEM with addition of Tetragonula sp honey and Apis mellifera royal jelly toward AMSCs proliferation. The research design used true experimental methods. Sample taken from a non-comorbid, healthy adult patient that underwent liposuction. AMSCs cultured with various concentrations of honey and royal jelly (0,05% and 0,1%), and the results of proliferation rates are determined with 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) assay or MTS assay. AMSCs cultured in 0,05% honey and royal jelly with additional 10% FBS has higher proliferation percentage than concentration groups of 0,1% with additional 10% FBS (p<0.05), 0,05% (p<0.05), 0,1% (p<0.05) and negative control (p<0.05). Nevertheless, the proliferation percentage still lower than positive control, which contain DMEM and 10% FBS without honey and royal jelly. Glucose composition in honey and royal jelly can inhibit MSCs proliferation, because of the formation of reactive oxygen species (ROS) induced by glucose excess. Further study about separate effects of honey and royal jelly usage as DMEM supplement and a safe amount of glucose threshold for AMSCs proliferation are needed, to make an optimal FBS alternative.
