Abstract

Non-alcoholic fatty liver disease (NAFLD) was an accumulation of intrahepatic lipids in hepatocytes without a history of excessive alcohol consumption, which can be exacerbated by oxidative stress. Parijoto fruit (Medinilla speciosa Blume) has been reported to contain various secondary metabolites, one of which is flavonoids that showed antioxidant activity to prevent oxidative stress. Optimization is needed to optimize the extraction of flavonoids and antioxidant activity from parijoto fruit to help reduce the exacerbation of NAFLD. This study aims to optimize total flavonoid content and antioxidant activity of parijoto fruit using Box-Behnken Design with variations in temperature, time, and extraction solvent concentration. TFC was determined using the complex formation with AlCl3 method and antioxidant activity was determined using the FRAP method. The optimal TFC was obtained at 80°C, 40 minutes, and solvent concentration of 96%, which was 9.67 mgQE/g extract. The optimal antioxidant activity was obtained at 80°C, 40 minutes, and solvent concentration of 96%, which was 129,0310 mgAAE/g extract. Optimal TFC and antioxidant activity of parijoto fruit extract were obtained at 80℃, 59,9974 minutes, and solvent concentration of 96% with TFC = 9,8183 ± 0,1135 mgQE/g extract and antioxidant activity = 123,8917 ± 0,0982 mgAAE/g extract within the 95% PI range, indicating that the extraction condition is valid and optimal.
