Abstract
Infertility remains a significant issue of the reproductive, with males contributing for approximately half of all cases. Diabetes mellitus is known to impair sperm quality through chronic hyperglycemia, which triggers increased oxidative stress. Excessive accumulation of reactive oxygen species (ROS) can damage sperm cell membranes, even induce DNA fragmentation, and disrupt cellular metabolism, ultimately leading to reduced sperm viability. Therefore, the administration of exogenous antioxidants is required to mitigate oxidative stress–induced damage. Pineapple core contains bioactive compounds such as flavonoid antioxidants and bromelain, which exhibit antioxidant activity and may serve as natural agents for protecting reproductive function This study uses a true experimental design using a post-test only control group approach. A total of 30 male Wistar rats were divided into five groups: a negative control group, a positive control group, and three treatment groups receiving pineapple core extract at concentrations of 50%, 75%, and 100%, respectively. Diabetes was induced using alloxan at a dose of 150 mg/kg body weight. Sperm viability was assessed using the eosin–nigrosin staining method. The results demonstrated significant differences in sperm viability between the positive control group and the treatment groups. An increase in sperm viability was observed with increasing concentrations of pineapple core extract, with the highest concentration (100%) producing the most pronounced effect. The improvement in sperm viability is presumed to be associated with the antioxidant activity of compounds present in pineapple core extract, which may reduce oxidative stress under diabetic conditions. Though the effects has a significancy value of P = 0,063. Overall, pineapple core extract shows potential as a natural antioxidant source for improving sperm viability in diabetic conditions.
