Abstract
Parijoto plants (Medinilla speciosa Blume) are known to contain bioactive compounds such as flavonoids, saponins, alkaloids, and tannins that have various pharmacological activities, but toxicity data are still limited. This research is an experimental research with true experimental design. The test animals used were female ddY mice with a minimum weight of 20 grams and 8-12 weeks of age. Leaf and fruit extracts were obtained through Ultrasonic Assisted Extraction (UAE) method with methanol solvent. Testing was carried out in two stages, namely preliminary tests, and fixed dose main tests at a dose of 2000 mg/kgBB. Parameters observed included toxicity symptoms, changes in body weight, relative weight of organs, as well as macroscopic and histopathological examination of liver and kidney organs. The results showed that the administration of parijoto fruit and leaf extracts did not cause death in test animals and only caused mild toxic symptoms in animals given fruit extracts. The LD50 value of both extracts was estimated to be more than 2000 mg/kgBB and classified as unclassified or non-toxic for leaf extract and category 5 or mildly toxic for fruit extract according to OECD. Histopathological examination showed mild changes in cellular structures, but not statistically significant. Thus, parijoto fruit and leaf extracts are relatively safe in acute oral administration at doses up to 2000 mg/kgBB.
