Abstract
Bacillary dysentery is one of the infectious gastrointestinal diseases caused by Shigella sonnei, with increasing levels of antibiotic resistance, particularly in developing countries. Parijoto (Medinilla speciosa B.) is an endemic Indonesian plant traditionally used by local communities as a herbal and is known to possess potential antibacterial properties. This study aimed to evaluate the in vitro antibacterial activity of methanolic extracts of Parijoto leaves and fruits against Shigella sonnei. Extraction was carried out using the Ultrasonic-Assisted Extraction (UAE) method with methanol as the solvent. Antibacterial activity was tested using the disk diffusion method to observe inhibition zones, and the broth dilution method was used to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), with extract concentrations ranging from 6.25 to 100 mg/mL. The results showed that both leaf and fruit extracts of Parijoto exhibited weak antibacterial activity against Shigella sonnei. The leaf extract showed its highest activity at a concentration of 25 mg/mL with a clear zone of 3.07 ± 0.12 mm, while the fruit extract demonstrated activity at 6.25 mg/mL with a clear zone of 1.7 ± 0.17 mm. However, MBC was not achieved for either extract. Statistical analysis revealed a significant difference (p < 0.05) in antibacterial activity between leaf and fruit extracts, with the leaf extract exhibiting greater effectiveness.
