Abstract
Hyperuricemia is a condition whose prevalence continues to increase globally and contributes to various pathological and chronic complications. With the potential of compounds it contains, melinjo seed extract (Gnetum gnemon L.) can be an alternative candidate for allopurinol,the aims is to determine the dose of extract that is most effective in reducing uric acid levels with graded doses (250, 500,1000 mg/kgBB). The research was experimental with pretest-posttest control group design. 30 rats were induced hyperuricemia using chicken liver juice and potassium oxonate for 7 days. Rats were randomly grouped into 5 groups (K-, K+, P1, P2, P3). Melinjo seed extract 250, 500, and 1000 mg/kgBB and allopurinol 10 mg/kgBB administered orally on the 8th day. Statistical analysis was performed to determine differences between groups before and after treatment. The findings reveal that the three doses significantly reduced uric acid levels (p<0.05) from 5.88 ± 0.54 to 4.64 ± 0.23; 5.98 ± 0.68 to 4.40 ± 0.29; 6.04 ± 0.59 to 3.92 ± 0.30. There is a significant difference between 250 and 1000 mg/kgBB. The effectiveness of the 1000 mg/kgBB dose reduced uric acid levels by 35%, 500 mg/kgBB (26%), 250 mg/kgBB (21%) and allopurinol (29%). In conclusion, Based on the result, melinjo seed extract can reduce uric acid levels with effectiveness equivalent to allopurinol. The most effective dose is 1000 mg/kgBW with a percentage reduction of 35% of the initial level.
