Abstract
Graptophyllum pictum (L.) Griff and Actinomycetes are natural resources known in Indonesia as medicinal plants and antimicrobial agents. G. pictum extract contains bioactive compounds with antibacterial properties, while Actinomycetes produce secondary metabolites effective against bacteria. This study evaluates and compares the antimicrobial effectiveness of G. pictum extract and Actinomycetes isolates extracted and isolated using the Ultrasound-Assisted Extraction (UAE) method against Staphylococcus aureus. The method used is a laboratory experiment with a comparative approach, involving the extraction of G. pictum and isolation of Actinomycetes, followed by antimicrobial activity testing using the well diffusion method. Results showed that G. pictum extract inhibited the growth of S. aureus at concentrations of 40%, 50%, and 60% with average inhibition zone diameters of 7.475 mm; 9.45 mm; and 10.05 mm; respectively. In comparison, Actinomycetes isolates showed average inhibition zone diameters of 5.35 mm at 40% concentration, 6.85 mm at 50% concentration, and 4.325 mm at 60% concentration. The Mann-Whitney test showed significant differences at 40% (p = 0.043) and 60% concentrations (p = 0.021), but not at 50% concentration (p = 0.248). The conclusion is that G. pictum extract has higher potential as an antimicrobial agent compared to Actinomycetes isolates against S. aureus. The UAE technique used in this study increases the efficiency of extracting active compounds from G. pictum and isolating Actinomycetes bacteria, resulting in higher concentrations of active compounds and secondary metabolites from both sources. It is hoped that this research can contribute to the development of alternative treatments for bacterial infections using natural materials.
