Abstract
Cumin leaf is potential to be developed as herbal medicine with largest content being flavonoids, extraction is required with influence of material to solvent ratio and extraction time. Toxicity tests are required to ensure safety of natural materials. The purpose of this research is to determine effect of material to solvent ratio and extraction time of cumin leaves on total flavonoid content and acute toxicity against Artemia salina. The method used is ultrasonic extraction, total flavonoid content assay with UV-Vis spectrophotometer and Brine Shrimp Lethality Test. The highest total flavonoid content was produced by extract with material to solvent ratio of 1:20 and 20 minutes with 28.5 mgQE/g extract. In the toxicity test, the most toxic LC50 value was obtained at material to solvent ratio 1:20 with 407.542. Based on the results of One-Way ANOVA test, the material to solvent ratio has significance value of less than 0.05 (p < 0.05) which affects total flavonoid content and the result of Pearson correlation test show a weak unidirectional relationship with toxicity of the extract, while extraction time does not significantly affect total flavonoid content and has a very weak directly proporsional correlation with toxicity of methanol extract of cumin leaves.
