Abstract

Stroke is a cerebrovascular disease that is a leading cause of death in Indonesia. Reduced blood flow in stroke can trigger excessive Reactive Oxygen Species (ROS) production and a significant decrease in antioxidant levels. Purple leaf (Graptophyllum pictum (L.) Griff) is a plant known for its phenolic compounds, which are recognized as a source of antioxidants for the body. Therefore, this study aims to analyze the effect of purple leaf extract on antioxidant levels in a rat model of ischemic stroke. The method used in this study is the True Experimental model. A total of 30 Sprague Dawley rats were divided into 5 control groups including those given citicoline, purple leaf extract at doses of 300 mg/kgBW, 150 mg/kgBW, and 75 mg/kgBW. The statistical test used in this study is One-Way Annova and the Post Hoc Bonferroni test. There were significant mean differences (p<0,05) in MDA, CAT, and GSH levels among treatment groups before treatment, 7th day of treatment, and 14th day of treatment. Based on the research results, it can be concluded that the best dose for increasing CAT levels is a dose of 75 mg/kgbb and a dose of 300 mg/ kgbb for GSH levels with the highest antioxidant levels on the day 7th of treatment then decreases on the 14th of treatment. Keywords: Antioxidant, Ischemic Stroke, Purple leaf.
