Abstract
Objective Breast cancer is a highly prevalent malignancy both in Indonesia and globally. The immune system plays a crucial role in tumor surveillance, particularly through cytotoxic T cells (CD8⁺), Natural Killer (NK) cells, and activating receptors such as NKG2D. This study aims to compare immune cell profiles from Peripheral Blood Mononuclear Cells (PBMC) between breast cancer patients without a history of chemotherapy and non-cancer individuals. Methods A retrospective analytic observational study was conducted using medical records from Hayandra Clinic (2020–2022). Immune cell subsets including CD3/CD8, CD56, CD19, and NKG2D were analyzed using Mann-Whitney test. Results Mann-Whitney analysis revealed that only the CD3⁻/CD8⁺ subset showed a statistically significant difference (p=0.013), with lower levels in breast cancer patients. Other subsets, such as CD3⁻/NKG2D⁺ and CD3⁺/NKG2D⁻, showed directional trends but were not statistically significant. Conclusion A significant decrease in CD3⁻/CD8⁺ cells indicate impaired cellular immunity in non chemoterapy breast cancer patients. These findings suggest that specific immune cell subsets may serve as potential biomarkers or therapeutic targets in breast cancer. Further studies with larger cohorts are recommended to validate these observations.
