Abstract
Background: Colon cancer remains a major cause of cancer morbidity and mortality, and cellular immunotherapy is being explored as an additional antitumor strategy. This study aimed to describe viability and apoptosis and analyze BCL-2 and P53 gene expression in HT-29 colon cancer cells after cytotoxic T cell treatment. Methods: This true experimental in vitro study used HT-29 cells as target cells and cytotoxic T cells isolated from cryopreserved PBMC-derived samples as effector cells. HT-29 cells were co-cultured with cytotoxic T cells at effector:target ratios of 10:1, 20:1, and 50:1 for 72 hours. Viability and apoptosis were assessed using Annexin V/PI flow cytometry, immune cell parameters were assessed by flow cytometry, and BCL-2 and P53 expression was measured using qRT-PCR with GAPDH as the reference gene. Results: Cell viability decreased from 91.10% in control cells to 78.62% and 52.40% at ratios of 20:1 and 50:1, respectively. Late apoptosis increased from 4.73% in controls to 11.30% and 39.74% at ratios of 20:1 and 50:1. BCL-2 expression was significantly reduced in all treatment groups (p = 0.000), whereas P53 expression showed a non-significant increasing trend (p = 0.159). Conclusion: Cytotoxic T cells reduced HT-29 viability, increased apoptosis, and significantly downregulated BCL-2 expression, while P53 expression did not change significantly.
