Abstract
Objective: Obesity is a major risk factor for oxidative stress, often marked by a decrease in endogenous antioxidants such as Glutathione Sulf Hydryl (GSH). This study aimed to evaluate the effect of Moringa oleifera fruit extract on GSH levels in the liver of obese Sprague-Dawley rats (Rattus norvegicus). Methods: This was a true experimental study using a post-test only control group design. Thirty male rats were randomly divided into five groups: normal control, negative control (obese without treatment), positive control (obese with vitamin C 15 mg/kgBW), and two treatment groups receiving Moringa oleifera fruit extract at 500 mg/kgBW once and twice daily for 14 days. Obesity was induced through a high-fat diet over 49 days. GSH levels in liver tissue were measured using spectrophotometry. Results: One-way ANOVA analysis showed a statistically significant difference in GSH levels between groups (p < 0.05). The group treated with 500 mg/kgBW of Moringa oleifera extract once daily demonstrated increased GSH levels comparable to the normal control. In contrast, the group receiving the extract twice daily showed a decrease in GSH levels. Although vitamin C increased GSH levels, the effect was less pronounced than in the once-daily Moringa group. Conclusion: A daily dose of 500 mg/kgBW Moringa oleifera fruit extract effectively increases hepatic GSH levels in obese rats. Higher frequency dosing may reduce GSH, indicating a potential pro-oxidant effect at elevated doses.
