Abstract
Abnormal, rapid, and uncontrolled breast cell proliferation is a hallmark of breast cancer. Numerous cancer therapy approaches have been discovered; however, some of them have been linked to immunological dysregulation and reduced physical capacity, which may contribute to post-treatment disease recurrence and metastasis. Immunotherapy, on the other hand, has shown a strong ability to increase cancer patients' quality of life and overall survival rates. A subset of cytotoxic lymphocytes, natural killer (NK) cells are the main effector cells in innate immunity against cancer cell invasion and are essential to cancer immunotherapy strategies. This study employs a true experimental design involving the co- culture of the MDA-MB-231 breast cancer cell line and NK cells, utilizing control and treatment groups with diverse effector:target (E:T) ratios (10:1, 20:1, 50:1), to assess cellular viability and apoptosis of the cancer cells utilizing flow cytometry and qRT-PCR. A p-value of less than 0.05 indicated a significant increase in the average and changes in BCL-2 gene levels. P53 gene levels, on the other hand, decreased on average and did not significantly alter (p-value > 0.05). Descriptive analysis revealed a decrease in viability and an increase in apoptosis, along with an increasing E:T ratio in the treatment group compared to the control group. Sel NK has been shown to significantly diminish cell viability and promote apoptosis, despite a rise in BCL-2 expression and a reduction in P53 expression, which cancer cells utilize as a mechanism to develop resistance to apoptosis.
