Abstract

Objective Antibiotic resistance has become a significant threat to global health, creating an urgent need for more effective new antibiotics. Secondary metabolites produced by endophytic bacteria are a potential source for novel antibiotic compounds. The stem of Portulaca grandiflora Hook. is known to contain secondary metabolites with antibacterial properties yet the potential of its endophytic bacteria remains underexplored. The purpose of this study is to isolate, identify, and evaluate the antibacterial activity of these endophytic bacteria against Escherichia coli and Staphylococcus aureus. Method This research is a true experimental study that begins with the isolation of endophytic bacteria from the stem of Portulaca grandiflora using the direct culture method, followed by bacterial identification through macroscopic and microscopic characterization and biochemical tests. The antibacterial activity was evaluated using Kirby–Bauer disk diffusion method. Result Three endophytic bacterial isolates (A, B, and C) were successfully obtained, all of which showed similarity to the genus Bacillus. Against E. coli, isolate B produced the largest inhibition zone (5,1 mm), classified as moderate, followed by isolate A (2,1 mm) and isolate C (1,3 mm), both classified as weak. Against S. aureus, isolate B also produced the largest inhibition zone (3,3 mm), followed by isolate A (1,5 mm) and isolate C (1,3 mm), all of which were categorized as weak. Conclusion The endophytic bacterial isolates obtained from the stem of P.grandiflora exhibited similarity to the genus Bacillus and demonstrated antibacterial activity against E.coli and S.aureus, classified as weak to moderate.
