Abstract
Objective Diabetes mellitus can reduce male reproductive quality through oxidative stress that impacts the decline in sperm motility, while pineapple stem (Ananas comosus (L.) Merr.) is known to contain antioxidant compounds that have the potential to protect sperm cells from oxidative damage. This study aims to determine the phytochemical content of pineapple stem extract, the success of diabetes induction in male Wistar strain rats with alloxan, and to test the effect of administering pineapple stem extract on sperm motility in diabetic rats. Method This study is experimental research with a post-test only control group design using male Wistar strain rats induced with alloxan to form a diabetes model. The rats were divided into a negative control group, a positive diabetic control group, and three treatment groups, each receiving 50%, 75%, and 100% concentrations of pineapple stem extract at 1 ml per day for a specified period, with evaluations of blood glucose levels, phytochemical tests of the extract, and microscopic assessment of sperm motility. The motility data were analyzed statistically using non-parametric tests adjusted to the data distribution to observe differences between groups. Result Treatment with pineapple stem extract at different concentrations did not produce a statistically significant difference in sperm motility (p>0.05). Nevertheless, descriptive analysis revealed a dose-dependent trend toward improved sperm motility with increasing extract concentrations. The group receiving the 100% extract exhibited the highest values of both progressive motility and total sperm motility among the experimental groups. Conclusion Pineapple stem extract demonstrated a tendency to improve sperm motility, with the most pronounced increase observed at the highest concentration (100%).
