Abstract

Fungal infections are a common health problem in Indonesia, a tropical country with high humidity. One of the most common fungal infections is dermatophytosis, an infection of keratinized tissue, and Trichophyton rubrum is the most common causative species. The use of synthetic antifungals such as ketoconazole carries the risk of side effects, so alternatives from natural sources are needed. Pandan leaves (Pandanus amaryllifolius roxb ) contain secondary metabolites such as flavonoids, tannins, alkaloids, phenolics, and steroids that have antifungal potential. This study aims to evaluate the effectiveness of pandan leaf ethanol extract against the growth of T. rubrum in vitro. The study was conducted as a true experimental study with a post-test only control group design. Extraction was performed using the ultrasound-assisted extraction (UAE) method with 96% ethanol solvent, then tested at five concentrations (40%, 50%, 60%, 70%, and 80%) using the well diffusion method on SDA media. Ketoconazole 2% was used as a positive control and distilled water as a negative control. The results showed that the ethanol extract of pandan leaves was able to inhibit the growth of T. rubrum at all concentrations tested, with a concentration of 80% producing the largest inhibition zone at 24 hours with an average size of 7,100 mm, which was categorized as moderate inhibition. Statistical analysis using the Kruskal –Wallis test showed significant differences (p < 0.05), followed by the Mann–Whitney test, which showed significant differences in most concentration comparisons (p < 0.05), except at concentrations of 40% and 50% at 48 hours of incubation (p > 0.05). The conclusion of this study is that ethanol extract of pandan leaves effectively inhibits the growth of T. rubrum, with a concentration of 80% being the most effective. This effectiveness is influenced by the content of secondary metabolites that damage the membrane and cell wall of the fungus. The decrease in the inhibition zone at 48 hours of incubation indicates a fungistatic property, where the extract only inhibits growth but does not permanently kill the fungus.
