Abstract
Alzheimer's disease is a neurodegenerative disorder characterized, in part, by a decrease in acetylcholine levels in the brain. Acetylcholinesterase (AChE) inhibitors play an important role in maintaining acetylcholine levels and are a primary therapeutic target for Alzheimer's disease. This study aimed to evaluate the in vitro AChE inhibitory activity of 70% ethanol extract and its fractions (96% ethanol, ethyl acetate, n-hexane) from andaliman fruit (Zanthoxylum acanthopodium DC.) Extraction was performed using maceration, followed by liquid–liquid fractionation. Analyses included qualitative phytochemical screening, determination of total phenolic content (TPC), AChE inhibition assay based on the Ellman method, and identification of active compounds using LC-HRMS. The ethyl acetate fraction showed the highest phenolic content (40.42 mgGAE/g) and the strongest AChE inhibitory activity (51.51%) with an IC50 value of 1784,73 ppm. ANOVA results showed a significant difference (p<0.05) in inhibition percentages among groups, while Pearson correlation analysis revealed a very strong positive correlation between AChE inhibition and total phenolic content ( p<0.001). LC HRMS analysis of the extract revealed the presence of alkaloids, flavonoids, coumarins, fatty acids, and aromatic amides. These findings indicate that Z. acanthopodium fruit has potential to be developed as a natural source of AChE inhibitors for the management of Alzheimer's disease.
