Abstract
Research on the expression of the Matrix Metalloproteinase 2 (MMP-2) gene shows a significant relationship with the effects of ultraviolet (UV) radiation, particularly in the context of skin aging (photoaging). Unlike many previous studies that utilized Human Dermal Fibroblasts (HDF), this research utilized leftover medical samples from palatal tissue, providing a new perspective on the effects of UV. Although sunlight contains more ultraviolet A (UVA), exposure to UVB is known to have a quicker damaging effect. Excessive UV exposure not only damages DNA but also affects proteins and lipids in cells, contributing to the aging process. In this study, four treatment groups were used: control group (0 minutes), 24 minutes, 48 minutes, and 96 minutes. UVB induction for each group was performed using a self-assembled device with a wavelength of 310 nm, which is known to increase oxidative stress through the formation of Reactive Oxygen Species (ROS). These ROS play a role in enhancing MMP-2 expression, which is crucial for the degradation of the Extracellular Matrix (ECM) and contributes to fibroblast aging. The results of the Kruskal-Wallis test showed a significance value (P-Value < 0.05), while the One-Way ANOVA indicated a significance value (P-Value > 0.05). Based on the research findings, it was concluded that UVB induction does not mediate the expression of the aging gene MMP-2 in fibroblast cells derived from oral mucosa.
