Abstract

Background : Infectious disease still being one of the main problems in Indonesia, mainly caused by pathogenic bacterium S. pyogenes and P. aeruginosa. Streptococcus pyogenes is a classical pathogens in human, essential in clinical cases, and cause local infections and sepsis in generalized and invasive infection. Pseudomonas aeruginosa is pathogenic bacteria major in clinical infection for causing high risk mortality due to the high risk bacteremia and the threat of antibiotics resistance. Black cumin (Nigella sativa, L.) has antimicrobial effect and contains active chemichal compound such as tannin, saponin, flavonoid, alkaloid, thymoquinone and dithymoquinone. Objective : This study aims to know effectiveness of black cumin extract against S. pyogenes and P. aeruginosa in-vitro. Method : This kind of research is experimental with a diffusion method, using Blood Agar for S. pyogenes and using Mueller Hinton Agar for P. aeruginosa. Result : Showed that the average of inhibition zone for S. pyogenes growth at concentration 20%, 40%, 60%, 80% and 100% are 4,51 ± 0,92 mm; 4,48 ± 0,27 mm; 6,21 ± 0,51 mm; 6,43 ± 0,73 mm and 7,33 ± 0,43 mm. Eventough, against P. aeruginosa growth at concentration 40%, 60%, 80% and 100% are 0,17 ± 0,12 mm; 0,86 ± 0,32 mm; 1,13 ± 0,23 mm and 1, 58 ± 0,58 mm. Kruskal-Wallis test (p < 0,05) showed that there was differences between each concentration for S. pyogenes and P. aeruginosa. Mann-Whitney test showed same effectiveness in some concentrations against S. pyogenes or P. aeruginosa. Conclusion : Black cummin (Nigella sativa, L.) extract has effectiveness for inhibition activity against S. pyogenes and P. aeruginosa.
