FACULTY OF MEDICINE PEMBANGUNAN NASIONAL “VETERAN” JAKARTA UNIVERSITY Undergraduate Thesis, June 2025 KENZA ANGGITA VINDASARI, No. NRP 2110211119 THE EFFECT OF DIFFERENT FREQUENCIES OF ULTRASOUND-ASSISTED EXTRACTION (UAE) ON THE EFFECTIVENESS OF Actinomycetes ISOLATES IN INHIBITING THE GROWTH OF Salmonella typhi BACTERIA PAGE DETAILS (68 pages, 8 tables, 3 figures, 11 appendices) ABSTRACT Objective Salmonella typhi is a Gram-negative bacterium that causes typhoid fever. Common treatment for this disease is the antibiotic chloramphenicol. Actinomycetes are known to be potential microorganisms because they can produce secondary metabolites that act as antibacterial agents. To obtain these antibacterial agents, extraction is necessary. Ultrasound-Assisted Extraction (UAE) is a modern extraction method that can significantly reduce work time and improve the yield and quality of the extract. Several factors are said to influence extraction results, including frequency. The purpose of this study was to determine the effect of different UAE frequencies at 30 kHz, 40 kHz, and 50 kHz on the effectiveness of Actinomycetes isolates as antibacterials in inhibiting the growth of S. typhi. Method The method used is purely experimental and conducted in vitro, or in a controlled environment. Antibacterial activity was tested using the well diffusion method, followed by measuring the inhibitory strength by observing the diameter of the clear zone formed. Result The results of this study showed the average diameters of the inhibition zones at extraction frequencies of 30 kHz, 40 kHz, and 50 kHz were 4,42 mm; 4,00 mm; and 5,76 mm, respectively. Data analysis using the One-Way ANOVA and Post Hoc test tests revealed significant differences in antibacterial inhibition among the treatment groups. Conclusion From the results of this study, it can be concluded that different frequencies of Ultrasound-Assisted Extraction (UAE) influence the effectiveness of Actinomycetes isolates in inhibiting the growth of Salmonella typhi.
