Abstract
Staphylococcus aureus is the main bacteria that causes various infectious diseases In humans, ranging from skin infections to bloodstream infections. Currently, S. aureus shows resistance to several antibiotics, prompting the search for natural treatments that do not cause resistance. Purple leaves (Graptophyllum pictum (L.) Griff.) are known in Indonesia as a herb that is used medicinally to cure hemorrhoids. This plant has potential as an antibacterial agent through its secondary metabolite compounds, such as flavonoids and tannins, which can be extracted using ultrasonic and maceration methods. This study aimed to compare purple leaf extract's antibacterial effectiveness between ultrasonic and maceration methods on the S. aureus growth. The experiment was conducted in vitro with a true experimental research design. Comparison of antibacterial abilities was carried out using purple leaf extract from ultrasonic and maceration methods at concentrations of 60%, 70% and 80%. The well diffusion method was used to test the antibacterial activity and inhibition zone measurements were carried out after 24 hours. The One-Way ANOVA test results revealed significant differences, and the Post Hoc Tukey test confirmed that the two methods at each concentration had a significant average difference compared to the negative control. There was a significant difference between the maceration and ultrasonic methods of extracting purple leaf, according to the independent sample t-test. In overall results, the ultrasonic extract of purple leaves showed a larger average zone of inhibition compared to the macerated extract. This advantage is due to the ability of the ultrasonic method to produce purer extracts without consuming a lot of energy and solvents.
